目的

探讨血红素加氧酶1(heme oxygenase, HO-1)过表达对人脊髓星形胶质细胞(human astrocytes of spinal cord, HA-sp)缺氧/复氧损伤的保护作用。

方法

通过给予HA-sp氯化钴(250 μl/L CoCl2)刺激模拟缺氧,然后再去除CoCl2刺激,建立细胞缺氧/复氧损伤模型。采用免疫印迹(western blot, WB)方法,从蛋白水平上检测不同刺激条件下HA-sp中HO-1的表达。采用四甲基偶氮唑蓝(methylthiazolyldiphenyl-tetrazolium bromide, MTT)方法,分析单纯给予缺氧/复氧刺激后以及上调HO-1表达的同时给予缺氧/复氧刺激后HA-sp的存活率变化。

结果

缺氧/复氧刺激条件下,HA-sp中HO-1蛋白过表达。利用锌原卟啉(ZnPPIX)抑制缺氧/复氧刺激条件下HO-1过表达后,HA-sp在缺氧/复氧刺激条件下存活率明显降低(P<0.01);与单纯给予缺氧/复氧刺激后HA-sp的存活率相比,用钴原卟啉(CoPP)上调HA-sp中HO-1表达的同时给予缺氧/复氧刺激,细胞存活率明显升高(P<0.01)。

结论

利用CoPP上调HA-sp中HO-1的表达,具有抗细胞缺氧/复氧损伤的保护作用。

Objective

To investigate the protective effect of overexpressed heme oxygenase-1(HO-1) in hypoxia-reoxygenation of human astrocytes of spinal cord(HA-sp).

Methods

Hypoxia-reoxygenation mode was established by oxygen provision after CoCl2 stimulation. The expression of HO-1 under different stimulation was detected by Western blotting. MTT assay was used to examine the relative survival of cell under hypoxia-reoxygenation as well as after up-regulated HO-1.

Results

HO-1 was upregulated by hypoxia/re-oxygen stimulation. The relative survival rate of HA-sp under hypoxia-reoxygenation was significantly decreased after HO-1 protein overexpression was inhibited by ZnPPIX (P<0.01), while the relative survival rate of HA-sp which was pretreated by CoPP was significantly increased compared with that of cells under hypoxia-reoxygenation (P<0.01).

Conclusion

HO-1 overexpression by CoPP exerts protective effect on hypoxia-reoxygenation injury of HA-sp. The result of this study may offer new thought for the therapy of hypoxia-reoxygenation injury of human spinal cord in the future.
